Combined Analysis of Whole Genome Re-sequencing and Transcriptome of Wheat Under the Stress of Tilletia controversa Kuhn
In order to reveal the difference of genetic background between susceptible cultivar(Yili053)and resistant cultivar(Zhongmai175)under the stress of Tilletia controversa Kuhn(TCK),and to screen the candidate genes of resistance to dwarf bunt,the study used the combined analysis of whole genome re-sequencing and tran-scriptom of kernels on two wheat cultivars,which were inoculated by TCK to evaluate the effect.After dif-ferential InDel analysis,a total of69 840 mutant genes were analyzed with transcriptome data,including 7 300 genes were expressed in wheat,and 162(2.22%)of the genes were significantly differential expressed,the fold change by more than 1.5 times(125 up-regulated and 37 down-regulated for'Zhongmai175'compared with Yili053);After differential SNP analysis,a total of 98 331 mutant genes were analyzed with transcriptome data,including 7 438 genes were expressed in wheat,and 126(1.69%)of the genes were significantly differential expressed,the fold change by more than 1.5 times(86 up-regulated and 40 down-regulated for'Zhongmai175'compared with Yili053).The genetic differential expression information and the genes with significant variation were successfully screened out,which lays a foundation for the further work of screening candidate genes for resistanceto dwarf bunt in wheat.The results might provide a scientific basis for disease resistance breeding of wheat in Xinjiang Uygur Autonomous Region.
