Cloning and Expression Analysis of JmDELLA Gene Related to Juglans mandshurica Hormone Regulation
In order to explore the regulation mechanism of hormone regulation related JmDELLA gene in the process of flower bud gender differentiation,the differential fragments of hormone regulation related DELLA gene were screened from the transcriptome sequencing database of Juglans mandshurica,and the full-length CDS sequence of DELLA gene was obtained by PCR amplification,named JmDELLA(GenBank accession number:400411).Bioinformatics software was used to bioinformatics analyze JmDELLA gene,and RT-PCR was used to analyze the expression of different organs and tissues of Juglans mandshurica.The total length of the CDs sequence of JmDELLA gene was 1 842 bp in length and encoding 613 amino acids.The results of amino acid sequence alignment and cluster analysis showed that the gene had more than 80% homology with Juglans regia,Pecan(Carya illinoinensis)and bayberry(Morella rubra).RT-PCR expression analysis showed that the JmDELLA gene related to the hormone regulation of Juglans mandshurica has the highest expression in female and male flower buds.Therefore,it is speculated that this gene is involved in the whole process of sexual differentiation of Juglans mandshurica flower buds.This result provides a theoretical basis for in-depth analysis of the molecular regulation mechanism of the heterogeneous sex differentiation of Juglans mandshurica.
