Development and Utilization of KASP Marker for Salt Tolerance Gene SKC1 in Rice
SKC1NB allele in indica rice variety'Nona Bokra',is a major QTL for salt-tolerant in rice.In order to im-prove the genotype screening efficiency for SKC1 locus,a KASP marker SKC1NB-KASP was developed based on the functional SNP site of the SKC1NB allele.Genotyping of 3 backcross population,which constructed with'Nona Bokra'as donor parent and Fanl4,HuaB and WulB as recurrent parent,respectively,indicated that among the 65 DNA samples of BC3F2 population,16 samples had the same genotype as the donor parent'Nona Bokra',31 sam-ples showed heterozygous genotype,and 18 samples had the same genotype as the recurrent parents.Subsequently,the SKC1 gene of these 65 DNA samples was sequenced to verify the accuracy of the KASP marker genotyping re-sults,and it was found that the sequencing results were completely consistent with the KASP marker genotyping results.These results indicate that the SKC1NB-KASP marker can efficiently and accurately screen genotype for SKC1 loci,and robust enough for molecular marker-assisted selection breeding for salt-tolerant rice.
