SSR标记对甜瓜杂交种'玉贵人'纯度的鉴定

为了精准快速地筛选出鉴定甜瓜杂交种纯度的SSR引物,本研究利用18对甜瓜SSR核心引物对甜瓜杂交种'玉贵人'及其亲本进行引物筛选和种子纯度鉴定.结果筛选出2对引物(CmS57和CmS59)在子代中兼具双亲条带并且在亲本间具有多态性,且条带清晰,间隔明显,并利用这2对引物在杂交群体中进行验证,纯度鉴定结果为97.8%和100.0%;SSR分子标记的鉴定结果与田间调查结果的误差为1.1%.表明本研究筛选出的引物CmS57和CmS59能够作为特异引物对甜瓜杂交种'玉贵人'进行种子纯度检测,也说明应用SSR分子标记技术对甜瓜种子的纯度鉴定是可行的.
Identification of Seed Purity of Melon Hybrid'Yuguiren'by SSR Molecular Markers
In order to accurately and quickly screen the SSR primers for identifying the purity of melon hybrids,in this study,18 pairs of melon SSR core primers were used to screen the primers and identify the seed purity of mel-on hybrids'Yuguiren'and its parents.The results showed that two pairs of primers(CmS57 and CmS59)had both biparental bands in the offspring and polymorphism among the parents with clear bands and obvious intervals,and were verified in the hybrid population with 97.8% and 100.0% purity;the error between the identification results of SSR markers and the field survey results was 1.1%.It showed that the primers CmS57 and CmS59 screened in this study could be used as specific primers for seed purity testing of melon hybrids'Yuguiren',and also indicated that the application of SSR molecular marker technology is feasible for the purity identification of melon seeds.
