Effects of Light Stress on the Metabolic Enzyme Activity of Polygonatum kingianum Coll.et Hemsl.
To study the effect of light stress on the polysaccharide content and metabolic enzyme activities in the root of Polygonatum kingianum Coll.et Hemsl.,different light condition were setted for P.kingianum growth us-ing a shading net.The polysaccharide content of rhizome and the activity of sucrose metabolism related enzymes[sucrose synthase(SS),sucrose phosphate synthase(SPS),sucrose neutral invertase(NI),sucrose acid invertase(AI)and uridine diphosphate glucose pyrophosphorylase(UGP)]were determined by spectrophotometric method.The results showed that the activity of sucrose metabolizing enzymes varied under different light stress.The NI and AI activities were the highest under 20%transmittance illumination and the SS,SPS and UGP activities were the highest under 100%transmittance illumination.With the increase of light intensity,the activities of SS,SPS and UGP increased,and the activities of NI and AI decreased.The correlation analysis between light intensity and five enzyme activities indicated that positively correlated was found between light intensity and SS activity.This study showed that light can affect the activity of sucrose metabolizing enzyme,and moderate light intensity is beneficial to the accumulation of rhizome.The reasonable regulation of light intensity has significant instruction to the accu-mulation of polysaccharides and the improvement of the quality of medicinal materials.The results provide a basic data and theoretical basis for the cultivation of P.kingianum and the cultivation of imitation field.
