Cloning and Bioinformatics Analysis of Pump Interaction Protein NTPpi1 Gene in Tobacco
In order to better select high potassium tobacco varieties,the gene sequence of NTPpi1 was cloned which based on the results of genome-wide association analysis of potassium content in tobacco leaves.Bioinformatics analysis showed that the length of NTPpi1 gene was 1 834 bp,the open reading frame was 981 bp,encoding a pro-tein sequence with 326 amino acids,and its molecular weight and isoelectric point were 145.3 kD and 5.00,respe-ctively.The online software TMHMM 2.0 Server predicted that there was a transmembrane helix structure with 23 amino acids.The amino acid sequences of the protein in different plants were aligned by DNAMAN software.The results showed that the amino acid sequence of the protein encoded by NTPpi1 gene was 80.82%consistent with that of other plants.Using NetPhos 3.1 Server software to predict the phosphorylation sites of NTPpi1,there were 29 phosphorylation sites were found,among which threonine phosphorylation site at 321 was located in the trans-membrane region.It is speculated that this site may be the site of interaction between the protein and ATPase pro-ton pump.These findings will provide theoretical support for the breeding of high potassium tobacco varieties.
