Germination Characteristics and Pollen Viability Comparison of Michelia shiluensis(Magnoliaceae)Stored Pollen
In this research,with pollens of Michelia shiluensis as test material,different concentrations of sucrose(50~150 g/L),H3BO3(100~300 mg/L)and CaCl2(200~400 mg/L)on pollen germination in vitro of Michelia shiluensis were observed.Moreover,the changes of pollen germination in vitro and stain rates of Michelia shiluen-sis at different storage temperatures(25 ℃,4 ℃,-20 ℃,-80 ℃)were studied.The results showed that:the medi-um with 50 g/L sucrose+100 mg/L H3BO3+400 mg/L CaCl2 was the most suitable for germination in vitro,and the importance of Michelia shiluensis pollen germination rate in vitro were H3BO3>sucrose>CaCl2 in order.Under dry conditions,-20 ℃ and-80 ℃,were more conducive to the preservation of Michelia shiluensis pollen.The MTT stain method was the most suitable for the rapid identification of the pollen activity of Michelia shiluensis and the magenta acetate stain method was not suitable for the determination of the pollen activity of Michelia shiluensis.This study can provide scientific and practical guidance for pollen collection,storage and vitality detec-tion of Michelia shiluensis.
