Induction of Cluster Buds and Establishment of Rapid Propagation Tech-nology in vitro of Fle mingia philip pine ns is Merr.et Rolfe
In order to optimize the best hormone conditions for each growth stage during the in vitro culture of Flemingia philippine ns is Merr.et Rolfe,to select the suitable explants and plant regeneration methods to establish a rapid propagation system.In this study,the stem segments,stem tips,and leaves of the vine-like aseptic seedlings with axillary buds were used as materials.The effects of different plant hormones and their concentrations on cal-lus induction,axillary bud germination,basal stem adventitious bud induction,and plant regeneration were studied through a complete combination experiment.The results showed that the proliferation mode of F.philippinensis was mainly direct organogenesis,and the stems with axillary buds were cultured in MS+6-BA 0.5 mg/L+NAA 0.2 mg/L+AC 1 g/L for 60 days.A large number of adventitious cluster buds grow,and the valueadded coefficient rea-ches 9.85;the suitable medium for rooting was MS+NAA 0.5 mg/L,the strong plant regeneration occurred 45 days later with the rooting rate up to 100%;the regenerated plants were transplanted to a matrix of peat soil∶humus soil∶perlite∶vermiculite=4∶4∶1∶1,and the survival rate reached 86%.In summary of the screened conditions,this study successfully established a fast propagation technique for the in vitro propagation of F.philippine ns is with good basal adventitious shoot induction and plant rooting effect and high survival rate of regenerated plants,which pro-vides an effective way to protect its wild resources and breed high-quality seedlings.
