地被菊CmbZIPs基因的生物信息及表达模式分析

地被菊(Chrysanthemum morifolium)为菊科菊属多年生草本花卉,其具有花朵繁密、花色丰富、抗逆性强等特点,是园林绿化美化的优良品种.土壤的盐渍化和低温严重限制地被菊的分布.植物中HD-ZIP基因家族广泛参与植物非生物胁迫.本研究以地被菊为材料,根据转录组数据克隆了 CmbZIP12、CmbZIP24、CmbZIP10、CmbZIP3基因,对其氨基酸序列、保守结构域等进行了生物信息学分析.在低温胁迫和NaCl胁迫处理下,利用RT-qPCR分析了这些基因的表达特性.结果表明,CmbZIP12、CmbZIP24、CmbZIP10、CmbZIP3基因ORF在453~630 bp之间,CmbZIP12和CmbZIP3为酸性蛋白,CmbZIP24和CmbZIP10为碱性蛋白,CmbZIP12、CmbZIP10和CmbZIP3为不稳定蛋白,CmbZIP24为稳定蛋白;利用MEME软件预测CmZIPs基序组成,共鉴定出4个保守基序;系统发育分析显示,CmbZIP12和CmbZIP3为HD-ZIPⅠ亚家族,CmbZIP10为HD-ZIPⅡ亚家族,CmbZIP24为HD-ZIPⅢ亚家族;互作网络预测发现CmbZIP12与GBF3、SNRK2.3、OST1等蛋白存在网络互作关系;CmbZIP24与AT5G04840、ZIP2、BZIP34等蛋白互作.低温和盐处理地被菊幼苗发现,CmbZIP12、CmbZIP24、CmbZIP10、CmbZIP3相对表达量都明显增加.本研究可为地被菊CmbZIP12、CmbZIP24、CmbZIP10、CmbZIP3 基因功能的研究提供参考.
Analysis of Biological Information and Expression Pattern of CmbZIPs Gene in Ground-cover Chrysanthemum
Ground-cover chrysanthemum(Chrysanthemum morifolium)is a perennial herbaceous flower of Dendran-thema genus in the family Compositae,in which has the characteristics of dense flowers,rich flower colors and strong stress resistance,and is an excellent variety for landscaping.However,the distribution of ground cover-chrysan-themum is restricted by low temperature and soil salinization.The HD-ZIP family is widely involved in abiotic stress in plants.In this study,CmbZIP12,CmbZIP24,CmbZIP10,CmbZIP3 genes were cloned from ground-cover chrysan-themum based on transcriptome data,and its amino acid sequence and conserved domain were analyzed by bioinfor-matics.Under low temperature and NaCl abiotic stress,the expression characteristics of these genes were analyzed by RT-qPCR.The results showed that the ORF lengths of CmbZIP12,CmbZIP24,CmbZIPI0,and CmbZIP3 genes were ranged from 453 to 630 bp.CmbZIP12 and CmbZIP3 were acidic proteins,and CmbZIP24 and CmbZIP10 were basic proteins;CmbZIP12,CmbZIP10 and CmbZIP3 were unstable proteins,and CmbZIP24 was a stable protein.Four conserved motifs were identified by MEME software.Phylogenetic analysis showed that CmbZIP12,CmbZIP3 belonged to HD-ZIPⅠ subfamily;CmbZIP10 and CmbZIP24 belong to HD-ZIPⅡ and HD-ZIPⅢ sub-family,respectively.Interaction network prediction showed that CmbZIP12 had network interaction with GBF3,SNRK2.3,OST1 and other proteins;CmbZIP24 interacted with AT5G04840,ZIP2,BZIP34 and other proteins.The relative expression levels of CmbZIP12,CmbZIP24,CmbZIP10 and CmbZIP3 were significantly increased by low temperature and salt treatment.This study can provide reference for the functions of genes CmbZIP12,Cmb-ZIP24,CmbZIP10 and CmbZIP3 in ground-cover chrysanthemum.
