新疆红肉苹果果肉花青苷积累及其相关基因表达

本研究采用pH示差法和实时荧光定量PCR探讨了 3个红肉苹果品种'夏红肉'、'红色之爱'、'克孜阿尔玛'在果实5个发育时期果肉中花青苷合成途径中其相关结构基因和转录因子的表达水平,以期为培育红肉苹果新品种提供理论参考.结果表明:'夏红肉'和'红色之爱'的果肉花青苷含量在幼果期和果实发育后期各有一个峰值;而'克孜阿尔玛'果肉花青苷随果实发育逐渐积累,至果实成熟时含量达到峰值;花青苷合成基因ANS、CHI和DFR的表达高峰在盛花后30 d或盛花后130 d和盛花后130 d,这与花青苷在不同积累型苹果中积累趋势相符.ANS、CHI和DFR是红肉苹果花青苷合成的关键结构基因,转录因子MYB10在3种新疆红肉苹果花青苷合成中起主导作用.
Accumulation of Anthocyanosides in the Pulp of Xinjiang Red-fleshed Apple and the Expression of Their Related Genes
In this study,we investigated the expression levels of anthocyanins and transcription factors in the flesh of three red apple varieties,'Summer Red Flesh','Red Love'and'Kizi Alma',during five developmental stages.We used the pH difference method and real-time fluorescence quantitative PCR to provide theoretical references for the cultivation of new red apple varieties.The results showed that the anthocyanin content of'Summer Red Flesh'and'Red Love'had a peak at the young fruit stage and one at the late fruit development stage,while the antho-cyanin content of Kizi Alma accumulated gradually with fruit development and reached a peak at fruit ripening;the expression peaks of anthocyanin synthesis genes ANS,CHI and DFR were 30 d or 130 d after flowering and 130 d after flowering.ANS,CHI and DFR are the key structural genes for anthocyanin synthesis in red flesh ap-ples,and MYB10 plays a dominant role in anthocyanin synthesis in the three Xinjiang red flesh apples.
