Identification,Phylogeny and Expression Analyses of DXS and DXR,Related to Isoprenoid Biosynthesis in Aquilaria sinensis
Essential oils of agar woods are fragrance source of agarwoods,a type of precious wooden plant.Isoprenoids are the main component of agarwood essential oils.Identification of genes responsible for biosynthesis of isoprenoids in agarwoods will help characterize molecular mechanisms of essential oil accumulation.This study focuses on DXS and DXR,two key genes in isoprenoid biosynthesis.The genes were identified by domain identification,mining of genes containing domains,and phylogenetic analysis.Results of domain identification revealed that both proteins of DXS and DXR contain 3 domains.Phylogenetic analysis showed that genes containing Transket_pyr domain can be classified into 6 major clades,while genes containing XP_redisom_C domain only has 1 clade.Through comparison with DXS and DXR genes in Arabidopsis,2 DXS genes and 1 DXR gene was identified from Aquilaria sinensis,respectively.Gene expression analysis showed that DXS genes were highly expressed in stems,leaves,flowers and flower buds,while DXR gene was putatively highly expressed.These results indicate that DXS is more likely a key gene for isoprenoid biosynthesis in agarwoods.The DXS and DXR genes identified in this study will provide basis for regulation of essential oil contents in agarwoods,and related results will produce theoretic value for essential oil component alteration in agarwoods.
