Transcriptome Sequencing and Bioinformatics Analysis of Trigonella foenum-graceum
Transcriptome sequencing and bioinformatics analysis of Trigonella foenum-graceum were performed based on RNA-Seq technology.The transcriptome data of Trigonella foenum-graceum were obtained to provide reference for future studies on the gene function,metabolic pathway,molecular marker and regulatory mechanism of Trigonella foenum-graceum.A total of 16 456 unigenes with a total length of 19 770 394 bp were obtained from the transcriptome data of Trigonella foenum-graceum.By comparing the obtained unigenes with seven databases,such as KEGG,Nr,GO and KOG,11 768 unigenes were annotated,accounting for 71.62%of the total unigenes.9 029 unigenes were annotated in GO database and divided into 3 categories and 52 subcategories according to their functions.There were 8 158 annotated unigenes in KEGG database,which were involved in 300 metabolic pathways.In addition,11 658 unigene were annotated in the Nr database,and the annotated homologous sequences were mainly from Medic ago truncatula.In KOG database,6 717 unigenes were annotated into 25 different functional classifications.A total of 11 286 SNP loci were identificated from the transcriptome data,among which C:G>T:A and T:A>C:G were the highest frequent mutation types.A total of 4 928 SSR loci were detected,and the single nucleotide repeats(2 262)accounted for 45.90%of the total.In this study,Trigonella foenum-graceum transcriptome database was constructed and bioinformatics analysis was performed,which provided scientific basis for identification of germplasm resources,analysis of genetic diversity and construction of fingerprint map of Trigonella foenum-graceum.
