摘要

磷是植物体的重要组成元素,也是植物生长发育所必需的大量元素之一,当前土壤的低磷胁迫严重制约着中国油菜和其他作物的生产.脂质转移蛋白LTPs(Lipid transfer proteins)是植物体中一类重要小分子量碱性蛋白,在油菜的抗低磷胁迫中有着潜在的应用价值.本研究从甘蓝型油菜中克隆了脂质转移蛋白基因BnLTP_C01,生物信息学分析显示该基因全长411 bp,能够编码含有136个氨基酸的蛋白质.该蛋白质第7~24个氨基酸的位置为跨膜区域,第53~135个氨基酸的位置为脂质转移蛋白结构域AAI.过表达载体2X35S::BnLTP_C01已构建完成,并成功遗传转化到甘蓝型油菜中,获得了 12个转基因阳性株系,为深入研究C01在油菜抗低磷胁迫中的作用提供实验材料,也为油菜抗低磷胁迫的分子育种提供一定理论基础.

Abstract

Phosphorus,an important component of plant body,is essential for plant growth and development.Cur-rently,low phosphorus stress of soil restricts the production of rapeseed and other crops seriously.As important basic proteins with small molecular weight in plants,LTPs(lipid transfer proteins)have potential utilization value in the resistance to low phosphorus stress in rapeseed.In this study,the gene BnLTP_C01 was cloned from Brassica napus.Bioinformatics analysis showed that BnLTP_C01 contained 411 base pairs,encoding a protein containing 136 amino acids.The transmembrane region of this protein is located at amino acids 7 to 24,and the lipid transfer protein domain AAI is located at amino acids 53 to 135.The overexpression vector 2X35S::BnLTP_C01 had been constructed and successfully transformed into Brassica napus.Twelve transgenic positive lines of Brassica napus were obtained,providing experimental materials to study on the function of BnLTP_C01 in rapeseed resistance to low phos-phorus stress.It also provides theoretical basis for molecular breeding of rapeseed resistance to low phosphorus stress.
