Grading Standard of Virus-free Tissue Culture Seedling of'Yujuhua 2 Hao'
Taking the stem of'Yujuhua 2 Hao'as the test material,tissue culture and detoxification treatment were studied to establish a rapid breeding system for'Yujuhua 2 Hao'chrysanthemum seedlings.The plant height and ground stem were determined as quantitative indicators for the classification.The results showed that the best ster-ilization plan for the stem of'Yujuhua 2 Hao'was soaked in 84 disinfectant solution(3.4％～4.6％chlorine)for 25 min;the best stem tip inducing hormone ratio was 6-BA 1.5 mg/L+NAA 0.1 mg/L.Through cluster analysis,'Yujuhua 2 Hao'tissue cultured seedlings were divided into level Ⅰ,level Ⅱ and level Ⅲ.The grading criteria:the grade stan-dard for Grade Ⅰ tissue culture seedlings was not less than 7.5 cm in plant height and ground stem was not less than 1.75 mm,a total of 11 plants.The grade standard for Grade Ⅱ was between 5.0～7.5 cm in plant height and 1.50～1.75 mm in stem,a total of 52 plants.The grade standard for Grade Ⅲ tissue culture seedlings was 2.5～5.0 cm in plant height and less than 1.50 mm in stem,a total of 27 plants,conform to normal distribution.This study pro-vides a reference basis for the promotion and production of'Yujuhua 2'variety.
