摘要

以'豫菊花2号'药食两用菊花茎段为材料,经过组织培养和脱毒处理,建立'豫菊花2号'菊花种苗快速繁育体系,确定株高和地茎为分级的量化指标.结果表明:'豫菊花2号'茎段的最佳灭菌方案为84消毒液(含氯量3.4%~4.6%)浸泡25 min;茎尖诱导最佳激素配比为6-BA 1.5 mg/L+NAA 0.1 mg/L;通过聚类分析将'豫菊花2号'脱毒苗分为Ⅰ级、Ⅱ级、Ⅲ级苗,分级标准:Ⅰ级脱毒苗的等级标准为株高不小于7.5 cm、地茎不小于1.75 mm,共11株;Ⅱ级脱毒苗的等级标准为株高在5.0~7.5 cm之间、地茎在1.50~1.75 mm之间,共52株;Ⅲ级脱毒苗的等级标准为株高在2.5~5.0 cm之间、地茎小于1.50 mm,共27株,符合正态分布.本研究为'豫菊花2号'品种推广生产提供参考依据.

Abstract

Taking the stem of'Yujuhua 2 Hao'as the test material,tissue culture and detoxification treatment were studied to establish a rapid breeding system for'Yujuhua 2 Hao'chrysanthemum seedlings.The plant height and ground stem were determined as quantitative indicators for the classification.The results showed that the best ster-ilization plan for the stem of'Yujuhua 2 Hao'was soaked in 84 disinfectant solution(3.4%~4.6%chlorine)for 25 min;the best stem tip inducing hormone ratio was 6-BA 1.5 mg/L+NAA 0.1 mg/L.Through cluster analysis,'Yujuhua 2 Hao'tissue cultured seedlings were divided into level Ⅰ,level Ⅱ and level Ⅲ.The grading criteria:the grade stan-dard for Grade Ⅰ tissue culture seedlings was not less than 7.5 cm in plant height and ground stem was not less than 1.75 mm,a total of 11 plants.The grade standard for Grade Ⅱ was between 5.0~7.5 cm in plant height and 1.50~1.75 mm in stem,a total of 52 plants.The grade standard for Grade Ⅲ tissue culture seedlings was 2.5~5.0 cm in plant height and less than 1.50 mm in stem,a total of 27 plants,conform to normal distribution.This study pro-vides a reference basis for the promotion and production of'Yujuhua 2'variety.
