摘 要TIFY是植物特有的转录因子,参与调控植物分生组织的分裂,能够减少机械损伤对植物造成的伤害,并且能提高植物的抗逆性。为了研究甘蓝型油菜中TIFY家族相关基因的功能,本研究采用同源克隆法在甘蓝型油菜品系1423和2205中克隆了 BnaA06g13250D的CDS序列,在NaHCO3胁迫后12、24 h,BnaA06g13250D基因编码在耐盐品系2205中的表达量显著低于敏盐品系1423,两亲本材料的耐盐性有明显差异。该基因CDS序列全长762 bp,编码253个氨基酸,含有一个TIFY结构域;Bna2205与甘蓝型油菜BnaA06g13250D基因的氨基酸序列存在16处差异,在第114个氨基酸序列处存在1处丝氨酸的增加,其蛋白质一级与二级结构也存在差异。系统进化分析表明,Bna2205基因与黑芥遗传距离最近,与番薯、植物棉、黑枸杞、山茶等不同属的物种遗传距离较远。利用blastp对甘蓝型油菜全基因组进行鉴定与筛选,得到了68个TIFY家族基因,都含有TIFY保守结构域,该基因家族包含3个亚家族,各亚族间在基因结构上存在显著差异,分布在19条染色体上。本研究为甘蓝型油菜TIFY家族相关基因的抗逆性研究提供理论基础,为甘蓝型油菜耐碱性盐的下一步研究提供参考。
Cloning of BnaJAZ1 Gene Related to Salt Tolerance in Brassica napus and Analysis of BnaTIFY Gene Family
TIFY is a plant-specific transcription factor that participates in the regulation of the division of plant meristems,which can reduce the damage caused by mechanical damage to plants,and can improve plant resistance to stress.In order to study the functions of the TIFY family related genes in B.napus,the homologous cloning method was used to clone the CDS(Coding sequence)of BnaA06g13250D in B.napus lines 1423 and 2205,at 12 and 24 h after NaHCO3 stress,the expression level of BnaA06g13250D gene in salt-tolerant line 2205 was significantly lower than that of salt-sensitive line 1423,and the salt tolerance of the two parent materials was significantly different.The results showed that the CDS of BnaJAZ1 was 762 bp,encoding 253 amino acids,and contained a TIFY domain;the Bna2205 cloned from the salt-tolerant variety 2205 had differences encoded by the BnaA06g13250D,there were 16 amino acid differences and one increase in amino acid sequence between Bna2205 and BnaA06g13250D gene of B.napus,and there were also differences in the primary and secondary structure of the protein.Phylogenetic analysis showed that the genetic distance between the BnaJAZ1 and B.nigra was the closest,but it was far from the species such as sweet potato.The study,we used blastp to identify and screen the whole genome of B.napus,and obtained 68 TIFY family genes,all of which contained TIFY conserved domains and three subfamilies,and there were significant differences in gene structure among the subfamilies,distributed on 19 chromosomes.This study laid a foundation for the research on the stress resistance of the TIFY family related genes,and provided a reference for the further study of alkali salt tolerance in B.napus.
