Construction of SSR Fingerprints and Genetic Diversity Analysis of 81 Clematis Cultivars
In order to reveal the genetic characteristics and explore the molecular identification methods of Clematis variety resources,the capillary electrophoresis detection with fluorescent SSR markers method was used to establish the DNA fingerprints and to analyze the genetic relationship of 81 Clematis varieties.The results showed that 10 pairs of SSR primers were screened out and amplified 129 alleles,with an average of 12 900 alle les;the average values fo effective number of alleles(Ne),observed heterozygosity(Ho)and expected heterozy-gosity(He)were 4.663,0.498 and 0.713,respectively.The Shannon's information index(I)ranged from 0.470~0.255,with an average of 0.687;the polymorphic information content(PIC)at different locus varied from 0.251~0.862 with an average of 0.687;the Nei's gene diversity index(H)ranged from 0.296~0.880 with an average of 0.713,suggesting that there is a high level of polymorphism in Clematis variety resources.Further,the result of NJ cluster analysis based on genetic distance showed that 81 Clematis germplasms were divided into four groups,which was highly consistent with the results of principal coordinate analysis(PCoA)and population genetic struc-ture analysis.Additionally,DNA fingerprints of 81 Clematis varieties were constructed by 10 pairs of SSR primers.The results of this study provide theoretical basis for the classification,identification,preservation and utilization of clematis species resources.
