Effect of Exogenous Abscisic Acid on Biosynthesis of Alkaloids in Uncaria rhynchophylla
The objective of this study was to investigate the effects of external application of ABA on the synthe-sis and accumulation of the active ingredients:rhynchophylline(RIN)and isorhynchophylline(IRN),as well as the gene expression of alkaloid synthesis pathways,in order to further reveal the biosynthesis of RIN and IRN in re-sponse to ABA mechanism.In this study,6-month-old Uncaria rhynchophylla was applied with different concen-trations of ABA,and the accumulation of RIN and IRN in U.rhynchophylla leaves after treatment with different concentrations of ABA was measured by HPLC,and analyze some key enzyme expression by fluorescence quantita-tive PCR(RT-qPCR).The expression of key enzyme genes STR,TDC,SGD and G8H in the synthetic pathway.The re-sults showed that 100 μmol/L ABA solution was most suitable for promoting the synthesis and accumulation of al-kaloids in U.rhynchophylla leaves.The content of 2 alkaloids increased by 1-fold compared with the control after 2 days treatment.At the same time,qPCR showed that SGD,TDC,G8H and STR family enzyme genes were all up-regulated to varying degrees.Therefore,reasonable external application of ABA solution will help promote the synthesis and accumulation of the active ingredients of U.rhynchophylla,improve the economic benefits of U.rhynchophylla,provide a theoretical basis for the efficient and high-quality planting management of U.rhyn-chophylla,and also clarify the biosynthesis of U.rhynchophylla alkaloids laid the foundation for the molecular reg-ulation mechanism.
