Bioinformatics Analysis of UDP-Glucose:Flavanone 7-O-Glucosyltransferase(UF7GT)
UDP-glucose:flavanone 7-O-glucosyltransferase(UF7GT)catalyzes the final step of flavanone bio-synthesis to form flavanone-7-O-glucoside in the circus. In this study, the nucleic acid sequences and amino acid sequences of UF7GT from 43 species registered in GenBank were analyzed by bioinformatics methods, including the primary structure, secondary structure, tertiary structure, and phylogenetic tree, etc. The results showed that the open reading frames(ORF)of UF7GT genes were about 1 326~1 926 bp,encoded 440~500 amino acids, and the isoelectric point was all lower than 7.Further,the investigation found that UF7GT in these species without leader peptides and signal peptides in N-terminal,the polypeptide chain presented hydrophilic and most of these UF7GT proteins are structural stable;the UF7GT proteins have non-transmembrane structures but two transmembrane helical structures in Pueraria montana var.lobata.The subcellular localization analysis found that existing and working on the plasma membrane or cytoplasm;the main motifs of the predicted secondary structure of UF7GT are alpha-helix and random coil, which belong to the GT-B superfamily, and most of them are highly conserved in evolution. The research would provide the basis for further investigating the function and structure characteristics of UF7GT and the biosynthesis of flavanone glycosides in the future.
