Microstructure Observation and Hormone-related Differential Genes Analy-sis of the Date Palm Seedlings about Shoots and Roots
Date palm is one of the most important woody food crops in arid and semi-arid regions. The morpho-logical and microscopic structure of the shoot and root are different, while the transcription mechanism is unclear. In order to explore the mechanism of differences between the shoot and root of date palm seedlings, transcriptome sequencing was performed in this study, focusing on the analysis of genes related to auxin, cytokinin, abolic acid, gibberellin, salicylic acid and jasmonin in plant hormone signal transduction pathways. The results indicated that the morphological and microscopic structure of the shoot and root are different, the leaves have spherical meso-phyll cells, the stems have relatively dense nuclei, and the middle of the root is the stele.Transcriptome analysis revealed there are 7 101 differentially expressed genes,3 042 upregulated and 4 059 downregulated.KEGG path-way analysis of differentially expressed genes showed that the second pathway was a plant hormone signal trans-duction.Genes of auxin TIR1,gibbecellins GID1,DELLA,TF,and salicylic acid NPR1 and PR-1 were highly ex-pressed in shoot. However, the genes of auxin AUX1 and PIN,cytokinin AHP,B-ARR,A-ARR and BIN3 genes, salicylic acid TGA,jasmonic acid JAZ,abscisic acid PP2C,ABF and SnRK2 genes were highly expressed in root. The plant hormone-related genes in shoot and root of date palm seedlings have tissue-specific expression, and the mechanisms involved in the regulation of the development of date palm seedlings are different, which provides the basis for the subsequent regulation of the growth and development of shoot and root of date palm seedlings by ex-ogenous hormones.
