Chloroplast Genome Characteristics of Pinellia cordata Revealed by Sequencing and Comparative Analyses with Related Species
To clarify the size,characteristics of the chloroplast genome in Pinellia cordata and the relationship of related species,this study sequenced and assembled the complete chloroplast genome of P.cordata.The chloro-plast genome(164 850 bp)is a typical double-stranded circular quadripartite structure,with 36.5%GC;its includ-ing a large single copy(LSC,89 964 bp),a small single copy(SSC,25 120 bp)and a pair of reverse repeats(IRs,24 883 bp);the whole chloroplast genome encoded 113 unique genes,including 79 protein-coding,30 tRNA and 4 rRNA genes;a total of 331 simple sequence repeats(SSR)were identified,and most of which had A/T base bias.Based on comparative analysis with chloroplast genome of seven Araceae,highly conservation in genome structure,gene order,and gene content were showed.The results of the contraction and expansion of IR regions at the junc-tion analysis that there was only one copy of trnN-GUU gene and a specific IR contraction.Consequently,the branch length of P.cordatta was longer than other species in the Pinellia monophyletic groups revealed by phylogenetic trees.This study provided the chloroplast genome sequence information of Araceae plants and indicated that P.cordata should possess special evolutionary history in comparison to other species in Pinellia.
