Effect of Overexpression of MeERF72 on Synthesis of C3 Cassava Starch
Cassava sucrose synthase 1(MeSuSyl)is the first key enzyme in the starch biosynthesis pathway,and ethylene response factor MeERF72 regulates its transcriptional activity.In order to investigate the effect of Me-ERF72 on the synthesis of starch in cassava(Manihot esculenta Crantz)root tubers,the expression levels of MeERF72 and MeSuSyl in leaves and tubers of wild type,MeERF72 overexpression lines and RNAi interfering lines were detected in pots,and the contents of starch,reducing sugar and total sugar in cassava root tubers were measured.The results showed that the expression level of MeERF72 in root tubers of overexpressed strains was significantly higher than that of wild-type C3,and the expression level of MeSuSyl in root tubers of overexpressed strains was significantly lower than that of wild-type C3,versa for interference strain;the content of starch and total sugar in potato meat of MeERF72 interference strain was significantly higher than that of wild-type C3,versa for overex-pression.These indicated that MeERF72 may negatively regulate the transcriptional activity of MeSuSy1,reduce sucrose decomposition efficiency,thus reduce starch synthesis.This study clarifies the regulatory mechanism of MeERF72 on MeSuSy1 and its effect on cassava starch synthesis,which will provide a reference for further re-search on the gene regulatory network of starch metabolic pathways and genetic improvement strategies for impor-tant traits in tropical crops.
