The Variety Purity Identification Based on SSR Markers of Watermelon New Hybrid Cultivars'Heijin'and'Shenmi968'
In this study,the watermelon hybrids cultivars'Heijin'and'Shenmi968',as well as their parent germ-plasms,were used as materials,the SSR molecular markers for the purify identification of the hybrid seeds were screened based on the agarose gel electrophoresis.Two primer pairs P10(BVWS00106)and P15(BVWS00433)were selected from 24 pairs of SSR core primers,for identification of hybrid cultivars'Heijin'and'Shenmi968',respec-tively.The specific PCR product bands of the hybrids and their parents were able to be clearly distinguished using 3%(w/v)agarose gel electrophoresis.The results showed the purity of'Heijin'and'Shenmi968'by the SSR marker,were 98.3%and 96.7%,respectively,and this result was highly consistent with the phenotypic characteristics of the plant in the field.The genetic diversity of the parent germplasms of these two hybrid cultivars were investigat-ed using SSR molecular markers,and constructed the fingerprint of parent germplasms,which provided technical support for the purity indoor rapid identification and the new cultivar protection of'Heijin'and'Shenmi968'.
