Analysis on Pathogenic Difference and Genetic Diversity of Leaf Stripe Pathogen in Hulless Barley
Stripe disease is one of the important seed-bome fungal diseases,which can cause serious yield loss to hulless barley production.In order to clarify the pathogenic variation and genetic diversity of hulless barley stripe pathogens distributed in Qinghai,this study compared the morphological characteristics of 18 stripe isolates from different areas in Qinghai Province,analyzed the genetic diversity among isolates by rDNA-ITS sequence and de-termined the pathogenicity variation of the pathogens.The color characteristics of the colony of the hulless barley stripe pathogen on the PDA medium were variable,and the looseness of the colony texture and the regularity of the edge had certain differences.Conidia cannot be produced on PDA medium;the colony diameter of the isolate ranges from 27.4 to 81.1 mm after the strain was cultured for 7 days,there were significant differences in the growth rates of the isolates.The results of pathogenic differentiation showed that there were differences in the pathogenicity of striped pathogenic isolates to hulless barley varieties.The 18 tested strains could be divided into 2 groups,and the pathogenicity had no obvious correlation with the regional source of the pathogen.According to the phylogenetic tree analysis,the 18 strains were divided into 2 subgroups when the Euclidean distance was 9.58,indicating that the level of genetic diversity of striae was low.There was no obvious correlation between the phy-logeny between isolates and their source regions,illustrating that stripe pathogens had more exchanges in different regions.This study can provide a theoretical basis for controlling barley stripe disease and the selection of isolates in subsequent studies.
