Establishment of Tissue Culture and Plant Regeneration System of Stellaria dichotoma
Stellaria dichotoma is a famous medicinal plant.Using MS as the basic medium,the explants were ob-tained by aseptic culture of S.dichotoma.By adding different concentrations of plant hormones such as 6-benzy-laminopurine(6-BA),naphthylacetic acid(NAA),6-furfuryl aminopurine(KT),indole butyric acid(IBA),the op-timal hormone ratio for the induction of adventitious buds and roots of S.dichotoma was explored,and the seedling refining and transplanting experiments were carried out.The results showed that the disinfection effect of S.dicho-toma treated with 15%NaCl for 10 min was the best,the pollution rate was only 3.37%,and the germination rate was 97.15%;the best medium for adventitious bud induction was MS+6-BA 1.5 mg/L+NAA 0 mg/L,and the induc-tion rate was 100%;the best medium for inducing adventitious bud rooting was 1/2 MS+6-BA 0.3 mg/L+IBA 0.5 mg/L,and the rooting rate could reach 90%;the seedlings were refined by half opening the cap of tissue culture bottle for 5 days and transplanted into matrix soil,and the survival rate was 100%.Using direct organogenesis,a system of tissue culture and plant regeneration of S.dichotoma was successfully established.These results will laid a foundation for the elite breeding and genetic improvement of S.dichotoma.
