涮辣与昆明皱皮椒pAMT生物信息学及表达分析

为分析涮辣与昆明皱皮椒假定的氨基转氨酶(pAMT)的差异,探究pAMT表达规律.本研究基于基因克隆和生物信息学方法分析pAMT基因和蛋白特性,利用qPCR测定pAMT相对表达量.结果显示,涮辣与昆明皱皮椒pAMT由1 380个碱基组成,编码459个氨基酸残基,相比昆明皱皮椒,涮辣pAMT存在7个同义突变和5个错义突变位点.顺式作用元件预测结果显示,辣椒pAMT可能受环境和植物激素的影响.蛋白特性结果显示,pAMT蛋白属于Ⅰ型折叠,Ⅲ类转氨酶,为脂溶性、亲水性的不稳定蛋白,主要定位在细胞质中,无信号肽与跨膜结构.两种辣椒pAMT蛋白配体结合位点、活性位点和蛋白磷酸化位点存在差异,不同物种AMT在进化上有保守性,但在亚组间也存在一定差别.在不同发育阶段,pAMT表达呈现先升高后急剧降低,且露地条件下pAMT表达量高于大棚;同一环境条件下,涮辣pAMT表达量整体高于昆明皱皮椒;不同外源物质在一定时间内影响pAMT的表达.相对于昆明皱皮椒,涮辣pAMT基因和蛋白的改变,可能是涮辣辣度极高的原因,并且不同生长环境、发育阶段和外源物质可能通过调节pAMT的表达来影响辣椒中辣椒素的生物合成.
Bioinformatics and Expression Analysis of pAMT in Shuanla and Kunming Wrinkled Pepper
In order to analyze the differences between Shuanla(Capsicum chinense)and Kunming wrinkled pepper(Capsicum annuum)pAMT enzymes and explore the changes of pAMT expression.The pAMT gene and protein char-acteristics were studied based on gene cloning and bioinformatics methods.The relative expression levels of pAMT were determined by qPCR.Shuanla and Kunming wrinkled pepper pAMT nucleotide sequences of 1 380 bp were ob-tained with ORF tools,which encoded 459 amino acids respectively.Compared with Kunming wrinkled pepper,Shuanla pAMT had 7 synonymic mutations and 5 sites of missense mutation.The cis-element prediction results illus-trated that pepper pAMT gene might be affected by environmental and plant hormones.Be predicted pAMT protein belonged to Ⅰ type folding,Ⅲ transaminase,as fat-soluble,hydrophilicity of unstable protein,mainly engaged in the cytoplasm,no signal peptide and the membrane structure.It was different for the ligand binding sites and active sites of pAMT.AMT of various species is evolutionarily conservative,but there had some differences among subgroups.The pAMT genes expressed that first increased and then sharply decreased under different development periods.It was higher for pAMT genes expression in the open field than greenhouses.Under the same environmental conditions,pAMT gene expression of Shuanla overall higher than Kunming wrinkled pepper.Different exogenous substances af-fect the expression of pAMT in a certain period of time.Compared with Kunming wrinkled pepper,changes in the pAMT gene and protein of Shuanla may be the reason for its high hotness.Capsaicin biosynthesis by regulating the ex-pression of pAMT in pepper under different growth environment,fruit development stages and exogenous substances.
