玉米PIN基因家族的鉴定和表达分析

生长素转运(PIN-FORMED,PIN)蛋白作为外向型运输蛋白质,在生长素的极性转运过程中发挥重要作用.为明确玉米(Zea mays L.)PIN基因家族的进化模式和对非生物胁迫的响应机制,本研究利用生物信息学方法对玉米PIN基因家族进行了特征分析.结果表明,玉米基因组中具有14个PIN基因家族成员,进化树中可分为4组.ZmPINs具有多样化的基因结构和较为保守的跨膜结构域.启动子分析结果显示,玉米PIN家族成员具有多因子植物激素响应元件和非生物胁迫元件,表明ZmPINs具有多样化的生物学活性.不同组织的数字表达谱分析表明,ZmPINs在不同阶段具有独特的调控作用.采用RT-qPCR技术检测了ZmPINs在非生物胁迫下的表达模式.结果显示,大多数ZmPINs在地上部受强烈诱导,在根中表达量受抑制.相反的表达模式说明,在非生物胁迫下,生长素在地上部和地下部之间存在动态运输过程.本研究可为明确玉米PIN基因家族在环境胁迫过程中的调控机制提供理论依据.
Identification and Expression Analysis of PIN Gene Family in Zea mays
PIN-FORMED(PIN)protein,as an outward transport protein,plays an important role in the polar trans-port of auxin.In order to clarify the evolution mode of the maize(Zea mays L.)PIN gene family and the response mech-anism to abiotic stress,this study used bioinformatics methods to analyze the characteristics of the maize PIN gene family.The results showed that there are 14 PIN gene family members in the maize genome,which can be divided into 4 groups in the evolutionary tree.ZmPINs have diverse gene structures and relatively conservative transmem-brane domains.The results of promoter analysis showed that members of the corn PIN family have multi-factor phytohormone response elements and abiotic stress elements,indicating that ZmPINs have diversified biological activities.The analysis of digital expression profiles of different tissues shows that ZmPINs have unique regulatory effects at different stages.The RT-qPCR technique was used to detect the expression pattern of ZmPINs under abi-otic stress.The results showed that most ZmPINs were strongly induced in the shoots and their expression in the roots was inhibited.The opposite expression pattern indicates that under abiotic stress,there is a dynamic transport process of auxin between the above-ground and underground parts.This study can provide a theoretical basis for clarifying the regulation mechanism of maize PIN gene family in the process of environmental stress.
