Construction and Verification of CRISPR/Cas9 Gene Editing Vector for Cassava MeXYLP1 Gene
Arabinogalactan proteins(AGP)is a glycoprotein widely distributed in various tissues and organs of plants,which actives an essential part in plant growth and development,especially in the formation of cell walls.Cassava MeXYLP1 gene is a member of a subfamily of AGP gene family.In order to identify its gene function,a target was designed on CRISPR-P v2.0 online website to construct gene editing vector pCAMBIA1301-Cas9-MeXYLP1-sg RNA.The cassava mutant of MeXYLP1 gene was induced by infecting cassava callus mediated by Agrobacterium tumefaciens.Sanger sequencing of gene editing sites and potential miss sites of transgenic calli showed that MeXYLP1 gene was successfully edited,and the potential miss sites were not missed.This study is helpful to obtain the cassava mutant of MeXYLP1 gene,so as to identify the function of its gene in the development of cassava cell wall.
