Transcriptome Analysis of Sarcopyramis nepalensis Wall.and Mining of Flavonoid Synthesis Related Genes
Sarcopyramis nepalensis Wall is a commonly used Chinese herbal medicine for protecting the liver and protecting the liver in southern Fujian.It is a natural medicinal plant resource with great medicinal,scientific and industrial development values.In this experiment,Illumina HiseqTM was used to sequence the transcriptome of the leaves and stems of SS.nepalensis,and Trinity software was used to complete the assembly of high-quality data transcripts of S.nepalensis,and then bioinformatics analysis was carried out.The research results showed that transcriptome sequencing was carried out on S.nepalensis leaf and stem samples,and each sample was repeated three times.A total of 262 444 416 high-quality reads were obtained.The N50 length was 734 bp,and the ave-rage length was 518.76 bp.The base percentage of Q20 was 98.65%~98.82%,and the base percentage of Q30 was 95.39%~95.88%.Then,seven important databases,including NR,GO,and KEGG,were used to compare them and obtain their functional annotation information,the results showed that 131 511 unigenes were annotated to at least one database,accounting for 61.44%;there are 530 differentially expressed unigenes in stem to leaf of S.nepalensis,of which 180 are upregulated and 350 are downregulated.In the analysis of differential genes between S.nepalensis leaves and stems,a KEGG database was used to analyze 112 significant KO pathways,and a total of 12 unigenes were found to be closely related to S.nepalensis flavonoid biosynthesis pathways.The TPM value of genes closely related to flavonoid biosynthesis in S.nepalensis transcriptome was extracted.Through this tran-scriptome sequencing study,a large number of S.nepalensis gene sequences were obtained,providing a reference basis for further mining the functional genes,biosynthesis regulation,quality improvement,etc.of the pharmaceu-tical active ingredients of S.nepalensis in the future.
