Cloning and Sequence Analysis of B-hordein Genes from Different Barley Varieties
Barley prolamins is one of the main factors affecting the food processing,malt production and feeding value.B-hordeins account for 70%~80%of whole prolamins and have great effects on barley quality.18 barley va-rieties from different countries and areas were used to clone the B-hordein genes,from which 27 complete gene se-quences were cloned,including 22 coding genes and 5 pseudogenes.By comparing with the sequences registered by GenBank,the base similarity between them was 79.6%~99.5%,indicating that all the 27 gene sequences were new genes or alleles.The deduced amino-acid sequences of 22 genes showed that 18 had a typical of 8 conserved cysteine(Cys)residues,2 genes contained 7 cysteine,and the remaining 2 genes contained 9 cysteine.The latter four genes were found the first time in barley,and might be the genes to code the good-quality subunits.The results above may provide a theoretical reference for further studying the characterization of B-hordein genes and breed-ing the barley varieties with good qualities.
