为了探寻最适合泰山白首乌新鲜叶片基因组DNA提取的方法,本研究以泰山白首乌新鲜叶片为材料,以5种不同的基因组提取方法提取泰山白首乌基因组DNA,并将提取到的基因组DNA经超微量分光光度计、凝胶电泳检测、聚合酶链式反应(PCR)检测,找出最适合提取泰山白首乌叶片基因组DNA的方法.在所有地方法中,CTAB法、改良CTAB法均可用于基因组提取,其中CTAB法提取的DNA浓度最高,凝胶条带单一清晰.本研究为泰山白首乌基因组高质量DNA的提取方法选择提供了参考,为后续进行分子生物实验提供科学依据.
Comparison of Genomic DNA Extraction Methods from Cynanchum bungei Decne.
For the purpose of exploring the most pertinent method of genomic DNA extraction from Cynanchum bungei,this study utilized the from fresh foliage of C.bungei the raw material,and gleaned the genomic DNA of C.bungei with five distinct extraction methods.Furthermore,the DNA was examined by ultra-micro spectrophotome-ter,gel electrophoresis,and polymerase chain reaction(PCR)testing to assessed and down-select the most appli-cable method for C.bungei DNA extraction.Among all the methods,CTAB method and improved CTAB method were proved to be suitable for genome extraction of this specific application,where CTAB method provided the highest concentration of DNA,with the proof of single and clear gel band result.This study provides future refer-ence for the selection of high-quality DNA extraction methods for C.bungei,and provides scientific evidence for subsequent experiments in the aspect of molecular biology.
