Development of Molecular Markers for Xanthomonas oryzae pv.oryzae Based on Genome Big Data
In order to screen the specific molecular markers for detecting Xanthomonas oryzae pv.oryzae(Xoo),this study was based on the 76 published Xoo,13 Xanthomonas oryzae pv.oryzicola(Xoc)and 30 peripheral micro-bial genomes data.Firstly,the bioinformatics analysis of three reported PCR(Polymerase chain reaction)molecu-lar markers of Xoo was firstly conducted.The results showed that among the DNA sequences of three PCR molec-ular markers,two markers had homologous sequences with Xoc,while one sequence was unique to Xoo.Addition-ally,there were three single nucleotide polymorphisms(SNPs)identified among the 76 Xoo strains.Then,four new specific DNA fragments existing only in the Xoo were screened by comparative genomics analysis.At last,five PCR molecular markers were designed to detect Xoo-infected leaf samples and Xoc-infected leaf samples.The re-sults showed that four molecular markers could only amplify the object band in the Xoo-infected leaf samples and no apparent bands in Xoc-infected leaf samples.This study established a method of molecular markers for Xoo based on genome big data,which enriched molecular markers for the identification of Xoo and references for the detection of other pathogens.
