Analysis of SSR Sequence Characteristics of Hordeum brevisubulatum Tran-scriptome
Hordeum brevisubulatum is a high-quality ecologically improved grass species,but currently there is little research on the development of molecular markers.Searching for SSR loci in 214 676 unigene sequences of H.brevisubulatum,a total of 24 877 SSRs were detected in 21 618 unigene sequences.The order of the number of repeat types was mononucleotide>trinucleotide>dinucleotide.The statistical analysis of the number of different re-peat types of H.brevisubulatum under different repeat times showed that the variation range of the repeat times was between 5 and 24 times,accounting for 65.70%,indicating that the development of SSR based on the tran-scriptome of H.brevisubulatum has a great possibility.Among the SSR repeat types,the number of SSRs with 5 repetitions was the largest,reaching 5 237,accounting for 21.05%of the total;followed by 10 repetitions,with a total of 4 861,accounting for 19.54%of the total.SSR primers were designed using Primer 3 software,20 802 primers were successfully designed out of the 24 877 SSR loci in the unigene sequence,with a success rate of 83.62%.Random detection of primers had a success rate of 80%.This study provides references for the develop-ment and application of SSR molecular markers of H.brevisubulatum,genetic evaluation of germplasm resources,and assisted breeding of molecular markers.
