Genetic Diversity of Ancient Lycium was Analyzed Based on matK,psbA-trnH,racL-a and trnL-trnF Sequences
Lycium is one of the important medicinal plants in Ningxia,and its nutritional value,economic value and ecological value are of great significance to Ningxia.Lycium ancient tree resources is one of the important germplasm resources in Lycium breeding.In this study,the genetic diversity of ancient Lycium barbarum was an-alyzed based on the sequences of chloroplast genes matK,psbA-trnH,racL-a and trnL-trnF.The results showed that the full length of the four genes was 2 892 bp,the conservative site was 2 863 bp,the mutation site was 29 bp,and the mutation rate was 0.01%.Taking Lycium L37 as an outgroup,a phylogenetic tree was constructed.The results showed that the genetic distance of all the ancient trees was small,indicating that the genetic years of the sample ancient trees were short and had not yet formed an independent population.In this study,four-segment chloroplast genes were used to analyze the genetic diversity of ancient Lycium trees,which provided a theoretical basis for subsequent introduction,domestication and breeding of Lycium,and was of great significance for the production practice of Lycium.
