Genetic Diversity Analysis and SSR Fingerprint Construction of Lonicera japonica and Its Related Species
In order to understand the differences of genetic background and genetic relationship between Lonicera japonica germplasm resources,in this study,genetic diversity analysis and fingerprint construction of 35 medicinal Lonicera japonica germplasm resources were carried out by SSR molecular marker technology.The results showed that the average number of effective alleles and expected heterozygosity of 23 pairs of primers were 2.891 and 0.618,respectively.The average Shannon information index and Nei's genetic diversity index were 1.300 and 0.609,respectively.The mean value of polymorphism information content of each site was 0.597,showing a high polymorphism.At the genetic similarity coefficient of 0.250,35 tested materials were divided into four categories.In addition,the fingerprint of Lonicera japonica germplasm resources was constructed by core primer method,and the rapid identification of Lonicera japonica and the related species at the molecular level was realized.This study provides a scientific basis for distinguishing genuine and fake Lonicera japonica,enriching excellent characters and breeding high-quality varieties.
