Secondary Metabolites of Endophytic Fungus Diaporthe sp.BMX12 from Aquilaria sinensis'Rekel'
To isolate the endophytic fungi from A quilaria sinensis'Reke 1'and study the fungal secondary metabo-lites.The endophytic fungi were isolated by tissue cutting method,and the species were identified by phylogenetic analysis of ITS sequences.The compounds of fermentation extract of endophytic fungi were isolated and purified by silica gel and Sephadex LH-20 column chromatography and high performance liquid chromatography(HPLC).The structures of all compounds were determined by spectroscopic methods including NMR,HRESIMS and ECD.Optical density method was used to determine the compounds'inhibitory activity against Staphylococcus aureus and Bacillus subtilis.The endophytic fungus Diaporthe sp.BMX12 was isolated and identified from Aquilaria sinensis'Reke 1'.Fourteen compounds,including one new compound,were isolated from its fermentation extract and identified as diaporthone H(1),diaporthone B(2),diaporthone D(3),cis-hydroxyscytalone(4),endoc rocin(5),4,6-dihydroxy-1(3H)-isobenzofuranone(6),monodictyphenone(7),diaporphasines A(8),citreorosein(9),4,6-dihydroxy-1H-isoindole-1,3(2H)-dione(10),1,3,8-trihydroxyanthraquinone(11),dicerandrol B(12),diceran drol A(13),12-O-deacetyl-phomoxanthone A(14).Compound 1 is a new polyketide.Compounds 12~14 show the inhibition rate over 50%on S.aureus ATCC 6538 and B.subtilis ATCC 6533.
