本研究以巴戟天为试验材料,探究试管苗生长的最适培养容器及离体保存的最适培养基,并以此为基础构建茎尖小滴玻璃化法超低温保存体系.结果表明,组培袋中植株长势更好且延缓生长方面较玻璃瓶和塑料瓶差异不显著,可作为巴戟天离体保存培养容器.高糖低激素培养基在巴戟天离体保存方面有显著效果,试管苗的保存时间从4个月延长至6个月.以高糖低激素培养基预处理的植株茎尖经过常温玻璃法小滴玻璃化法超低温保存后再生率可达13.3%.本研究结果为巴戟天离体保存和其茎尖小玻璃化超低温保存体系的建立奠定基础.
Construction of in Vitro Preservation and Shoot Tips Droplet-vitrification Cryopreservation System of Morinda officinalis How
In this study,the Morinda officinalis was used as the experimental material to explore the optimal cul-ture container for the growth of test-tube seedlings and the optimal medium for in vitro preservation.Based on this,the cryopreservation system of shoot tips droplet-vitrification was constructed.The results showed that the plant growth in the tissue culture bag was better and the growth delay was not significantly different from that in the glass bottle and the plastic bottle,which could be used as the in vitro preservation culture container of Morinda officinalis.The high sugar and low hormone medium has a significant effect on the preservation of Morinda officinalis in vitro.The preservation time of tissue culture seedlings was extended from 4 months to 6 months.The regeneration rate of shoot tips pretreated with high sugar and low hormone medium could reach 13.3%after cryopreservation by room temperature droplet-vitrification.The research results laid a foundation for the in vitro preservation of Morinda officinalis and for establishing its shoot tips droplet-vitrification cryopreservation system.
