Cloning and Genetic Transformation of Carotenoid Synthesis Gene MtSSUII in Banana
SSUII is a key gene in plant carotenoid synthesis pathway.By analyzing Karat transcriptome data of TTT genotype bananas rich in carotenoids,SSUII genes related to carotenoid synthesis in bananas were screened.The cDNA sequence of SSUII gene was cloned from Karat banana leaf tissue by PCR and sequenced successfully.The results showed that the open reading frame(ORF)of SSUII gene sequence was 1 053 bp,encoding 350 amino acids and the molecular weight was 37.10 kD,and the protein was hydrophobic and unstable.Through comparative analysis,it was found that the SSUII protein sequence had the conserved domain of the PTS family--the FARM domain rich in aspartic acid,named MtSSUII.The results of fluorescence quantitative PCR showed that the expression of MtSSUII gene was different in different genotypes of bananas during the same growth period,with TTT>BB>AAA.The relative expression levels of MtSSUII gene were different in different parts of Karat banana with TTT genotype during the same growth period.The expression levels were in the order of fruit>stem>flower>root>leaf from high to low.The 35S promoter detection of banana overexpressing transgenic plants obtained by genetic transformation showed that 4 plants were positive.The relative expression level of MtSSUII gene in transgenic positive plants was higher than that in non-transformed plants,and the content of carotenoid in transgenic positive plants was higher than that in control plants.In conclusion,it is preliminarily determined that MtSSUII gene plays a positive regulatory role in the pathway of banana carotenoid synthesis and promotes the synthesis of banana carotenoid.In this study,we explored the response mechanism of MtSSUII gene to carotenoid synthesis,and provided theoretical and technical reference for the cultivation of high carotenoid bananas.
