ABF(ABRE binding factors)转录因子是能够特异识别ABA响应元件(ABA-responsive element,ABRE)的一类碱性亮氨酸拉链蛋白,参与调控ABA响应基因的表达,在植物响应非生物胁迫的过程中具有重要作用。为获得蓝莓ABF转录因子基因序列,本研究通过同源比对和RT-PCR技术,从蓝莓品种'蓝丰'中克隆得到ABF转录因子基因VcABF2(GenBank登录号:MZ516811)。序列分析表明VcABF2基因全长为1 302 bp,编码377个氨基酸,相对分子质量为40。89 kD,理论等电点(pI)为9。46。生物信息学预测结果显示VcABF2蛋白定位于细胞核,为不稳定的亲水蛋白,含有bZIP转录因子家族保守结构域。系统发育分析结果表明,VcABF2蛋白与甜杨、拟南芥的ABF蛋白聚为一支。进一步以VcGAPDH为内参基因,实时荧光定量PCR结果显示,VcABF2受盐胁迫的诱导上调表达。本研究为探索VcABF2响应盐胁迫的分子机制提供了理论依据。
Cloning and Expression Analysis of ABF Transcription Factor Gene VcABF2 in Blueberry
ABF(ABRE binding factors)transcription factors are the basic leucine zipper protein that can specifi-cally recognize ABA-responsive elements(ABRE),involved in regulating the expression of ABA-responsive genes,which play important roles in the response of abiotic stress in plants.To obtain the blueberry ABF transcrip-tion factor gene sequence,the ABF transcription factor gene VcABF2(GenBank accession number:MZ516811)was cloned from the blueberry variety'Lanfeng'by homology comparison and RT-PCR technology.Sequence anal-ysis showed that the full length of the VcABF2 gene was 1 302 bp,encoding 377 amino acids,with a relative molecular mass of 40.89 kD and theoretical isoelectric point(pI)of 9.46.Bioinformatic predicted that VcABF2 protein was located in the nucleus and was an unstable hydrophilic protein containing conserved domains of the bZIP transcription factor family.From the phylogenetic analysis,the VcABF2 protein clustered with ABF proteins of Populus suaveolens and Arabidopsis thaliana.Furthermore,using VcGAPDH gene as an internal reference gene,the results of real-time quantitative PCR showed that the expression of VcABF2 was up induced by salt stress.This study provides a theoretical basis for exploring the molecular mechanism of VcABF2 in response to salt stress.
