Cloning and Expression Analysis of ABF Transcription Factor Gene VcABF2 in Blueberry
ABF(ABRE binding factors)transcription factors are the basic leucine zipper protein that can specifi-cally recognize ABA-responsive elements(ABRE),involved in regulating the expression of ABA-responsive genes,which play important roles in the response of abiotic stress in plants.To obtain the blueberry ABF transcrip-tion factor gene sequence,the ABF transcription factor gene VcABF2(GenBank accession number:MZ516811)was cloned from the blueberry variety'Lanfeng'by homology comparison and RT-PCR technology.Sequence anal-ysis showed that the full length of the VcABF2 gene was 1 302 bp,encoding 377 amino acids,with a relative molecular mass of 40.89 kD and theoretical isoelectric point(pI)of 9.46.Bioinformatic predicted that VcABF2 protein was located in the nucleus and was an unstable hydrophilic protein containing conserved domains of the bZIP transcription factor family.From the phylogenetic analysis,the VcABF2 protein clustered with ABF proteins of Populus suaveolens and Arabidopsis thaliana.Furthermore,using VcGAPDH gene as an internal reference gene,the results of real-time quantitative PCR showed that the expression of VcABF2 was up induced by salt stress.This study provides a theoretical basis for exploring the molecular mechanism of VcABF2 in response to salt stress.
