Construction of DNA Fingerprints of Chicory(Cichorium intybus L.)Germ-plasm Resources Based on SSR Markers
In order to excavate high grade chicory in domestic and foreign germplasm resources,supplement the DNA fingerprint of chicory germplasm resources,provide technical support for the selection of chicory breeding materials and identification of new varieties.In this study,using 23 chicory resources from different sources as ma-terials,SSR molecular marker technology was used to analyze the genetic diversity of 23 chicory and construct molecular fingerprints.The results showed that PCR amplification was performed in 50 pairs of SSR primers,and 8 pairs of primers with clear and stable bands and high polymorphism were screened,accounting for 16%of the primers used.The selected 8 pairs of SSR primers amplified a total of 82 bands in 23 chicory germplasm resources,with an average of 10.25 loci detected per pair of primers,of which 76 were differential bands,and the polymor-phism ratio was as high as 92.7%.The amplified fragment size ranged from 50 to 500 bp.On average,9.5 poly-morphic bands amplified by each primer,and the variation range was 6~11.The PIC value of the polymorphism information of 8 pairs of primers ranges from 0.33 to 0.60,with an average of 0.49.Using UPGMA method,23 chicory resources could be clustered into 5 categories at GS 0.53.Selecting three pairs of primer combinations of JJ13,JJ31 and JJ125 can accurately and quickly distinguish 23 chicory germplasm resources.The genetic diversity and fingerprint results of chicory germplasm resources in this study will provide technical support for the selection of chicory,breeding materials and the identification of new varieties.
