Screening of Primers for DNA Barcoding of Tea Plant
In order to screen out relevant DNA barcode primers needed for classification among different varieties of tea plants.In this study,the common primers of matK,T-ITS,trnH-psbA from other literatures and the self-de-signed rbcL primers were used to analyze the relevant sequence diversity of tea plants.The total DNA of the genome was extracted with the kit,the alternative primers were amplified by PCR,and the PCR amplification products were detected by agarose gel electrophoresis.The results showed that from the 5 pairs of primers matK,rcbL,T-ITS,trnH-psbA and rbcL,2 pairs of primers suitable for tea plant DNA barcode were preliminatively screened,namely matK and rcbL,which could be considered as alternative sequences for tea plant DNA barcode through analysis.It provides some reference for the study of tea variety classification and germplasm resources.
