漆树种质资源SSR遗传多样性分析

漆树是中国重要的经济树种.为了能够保护且合理开发利用漆树资源,本试验采用SSR分子标记技术,对来自陕西、甘肃、河南和云南4个省份的39个地区117个漆树样本进行遗传多样性分析和聚类分析.研究结果显示,10对漆树SSR引物均能对漆树样本进行PCR扩增且多态性丰富(A=9.400 0,PIC=0.714 6);漆树在物种水平上遗传多样性丰富(Na=1.828 2,I=0.553 2,H=0.393 7),种群间有明显的遗传分化(FST=0.375 4).UPGMA聚类分析显示相同种群的样本优先聚为一类,117份漆树样本明显聚为5个大类,表明SSR指纹图谱对漆树样本在种群水平上有良好的鉴定效果.本研究结果为漆树种质资源分类、鉴定和保护提供了重要依据.
Genetic Diversity Analysis of SSR Germplasm Resources of Toxicodendron vernicifluum
Toxicodendron vernicifluum(Stokes)F.A.Barkley is an important economic tree species in China.In order to protect and rationally utilize T.vernicifluum resources,This study utilized SSR molecular marker techno-logy to analyze the genetic diversity and perform cluster analysis on 117 samples from 39 regions of Shaanxi,Gansu,Henan and Yunnan.The results showed that all the 10 pairs of microsatellite primers could amplify the T.vernicifluum samples by PCR with rich polymorphism(A=9.400 0,PIC=0.714 6);there was high genetic diversity at the species level(Na=1.828 2,I=0.553 2,H=0.393 7),and there was a significant genetic differentiation among populations(Fsr=0.375 4).UPGMA cluster analysis showed that the samples from the same population always had priority clustering,and 117 samples of T.vernicifluum clustered into 5 large branches,indicating that SSR finger-printing had a good identification efficiency for T.vernicifluum samples at the population level.The results of this study provide an important basis for the classification,identification and protection of T.vernicifluum germplasm resources.
