摘要
目的:建立HPLC法测定藏荆芥挥发油中E,Z-荆芥内酯特征成分含量的方法.方法:以十八烷基硅烷键合硅胶为填充剂(色谱柱:Supersil ODS 250mm×4.6mm,5μm),以乙腈-水(40∶60)为HPLC的流动相,流速为1 mL/min,色谱柱的柱温为30 ℃,检测波长为216nm.结果:线性关系方程为y=1E+07x+287501,R2=0.9991,在0.0098~2.4405 mg/mL范围内的峰面积与进样浓度线性关系良好.精密度和重复性良好,峰面积RSD分别为0.66%、0.32%,加样回收率平均值为101.67%,24h内样品溶液稳定.结论:首次确认藏荆芥挥发油中含有E,Z-荆芥内酯,所建立的高效液相色谱法方法简单、准确、快速,重复性强,专属性好,可应用于藏荆芥E,Z-荆芥内酯特征成分的含量测定.
Abstract
Objective:To establish a method for the determination of E,Z-nepetalactone in volatile oil of Nepeta angustifolia C.Y.Wu by HPLC.Methods:Octadecylsilane bonded silica gel was used as filler(column:Supersil ODS 250 mm×4.6 mm,5 μm),acetonitrile-water(40∶60)as HPLC mobile phase,the flow rate was 1 mL/min,the column temperature was 30 ℃,and the detection wavelength was 216nm.Results:The linear relationship equation was y=1E+07x+287501,R2=0.9991,and the peak area in the range of 0.0098~2.4405 mg/mL had a good linear relationship with the injection concentration.The precision and repeatability were good,the RSD values of peak area were 0.66%and 0.32%,respectively,the average recovery rate was 101.67%,and the sample solution was stable within 24 hours.Conclusion:It was confirmed for the first time that the volatile oil of Nepeta angustifolia C.Y.Wu contains E,Z-nepetalactone.The established high-performance liquid chromatography method is simple,accurate,rapid,reproducible and specific,and can be applied to the determination of the characteristic components of E,Z-nepetalactone.
