UPLC测定不同牛大力样品中刺桐碱的含量

目的:优化牛大力中刺桐碱的提取方法 ,并建立测定牛大力中刺桐碱含量的超高效液相色谱(UPLC)方法,从而比较不同牛大力中刺桐碱的含量.方法:采用单因素和正交实验优化牛大力中刺桐碱的提取,含量测定采用Agilent XDB-C18(1.8 μm,2.1×100 mm)色谱柱,乙腈水溶液梯度洗脱,检测波长278 nm,流速0.3mL/min,柱温30 ℃.结果:确定牛大力中刺桐碱80℃水浴回流提取的最佳条件为料液比为1∶15g/mL,提取时间为1.5h,乙醇浓度为70%;建立的刺桐碱含量测定方法快速、准确、稳定、可靠.结论:22个不同的牛大力样品中刺桐碱含量差别较大,与品种、生长环境和生长年限等因素有关.
Determination of Hypaphorine in Roots of Millettia Speciosa Champ.by UPLC
Objective:To optimize the extraction method of Hypaphorine in Millettia speciosa Champ,and to establish an ultra performance liquid chromatographic(UPLC)method for the determination of Hypaphorine in Millettia speciosa Champ,so as to compare the contents of Hypaphorine in different Millettia speciosa Champ.Methods:The extraction of Hypaphorine from Millettia speciosa Champ was optimized by single-factor and orthogonal experiments.Agilent XDB-C18column(1.8 μm,2.1×100 mm)and the mobile phase was eluted with a gradient of acetonitrile-water solution.The detection wavelength was set at 278 nm and the flow rate was 0.3 mL/min.The column temperature was maintained at 30 ℃.Results:The optimum conditions for the reflux extraction of Hypaphorine from Millettia speciosa Champ at 80 ℃ were determined as the material-liquid ratio of 1∶15 g/mL,the extraction time of 1.5 h,and the concentration of ethanol of 70%;the established method for the determination of Hypaphorine was rapid,accurate,stable and reliable.Conclusion:The content of Hypaphorine in 22 different Millettia speciosa Champ samples varied widely and was related to factors such as species,growing environment and growing years.
