一锅法合成取代的3-碘-1-对甲苯磺酰基-1H-吲哚的反应研究

3-碘-1-对甲苯磺酰基-1H-吲哚是一类重要的合成反应中间体,在金属催化的偶联反应中应用广泛。本文以简单易得的各类吲哚为原料,在引入不同取代基的情况下,考察N-H被Ts基团保护的3-碘代吲哚的合成。具体为以N,N-二甲基甲酰胺为溶剂,氢氧化钾为碱,以各种基团取代的吲哚、碘与4-甲苯磺酰氯为原料一锅法制备目标产物。总计有17 个含有不同取代基的3-碘-1-对甲苯磺酰基-1H-吲哚被合成,结构经 1H-NMR分析确认。经一锅法合成取代的3-碘-1-对甲苯磺酰基-1H-吲哚在后处理过程中无需柱层析,简单洗涤即可得到纯度较高的目标产物,适用于工业化生产。
Synthesis of Substituted 3-Iodo-1-Tosyl-1H-Indole by One-Pot Method
3-iodo-1-tosyl-1H-indole is a very important intermediate in organic synthesis.It is widely used in metal-catalyzed coupling reactions in fact.In this paper,the synthesis of 3-iodoindole protected by Ts group in the presence of different substituents were studied.Specifically,The target product was prepared by using N,N-dimethylformamide as the solvent,potassium hydroxide as the base,substituted indoles,iodine,4-toluenesulfonyl chloride as raw materials in one port.As a result,seventeen 3-iodo-1-tosyl-1H-indole containing different substituents were synthesized and their structures were confirmed by 1H-NMR.Under the condition of introducing different substituents,the one-pot synthesis of substituted 3-iodo-1-tosyl-1H-indole can be achieved successfully.This method is simple to operate and the products were easy to be purified without the need for column chromatography.Therefore,it is very suitable for industrial production.
