Synthesis of Substituted 3-Iodo-1-Tosyl-1H-Indole by One-Pot Method
3-iodo-1-tosyl-1H-indole is a very important intermediate in organic synthesis. It is widely used in metal-catalyzed coupling reactions in fact.In this paper, the synthesis of 3-iodoindole protected by Ts group in the presence of different substituents were studied.Specifically,The target product was prepared by using N,N-dimethylformamide as the solvent, potassium hydroxide as the base, substituted indoles,iodine,4-toluenesulfonyl chloride as raw materials in one port.As a result, seventeen 3-iodo-1-tosyl-1H-indole containing different substituents were synthesized and their structures were confirmed by 1H-NMR.Under the condition of introducing different substituents, the one-pot synthesis of substituted 3-iodo-1-tosyl-1H-indole can be achieved successfully. This method is simple to operate and the products were easy to be purified without the need for column chromatography. Therefore,it is very suitable for industrial production.
