High Performance Liquid Chromatography-evaporative Light Scattering Detection Method to Determine the Component Content of Pentanediamine
A new analytical method for the detection of pentanediamine was developed by reverse liquid chromatography tandem evaporative light detector.The evaporative light detector has good versatility and overcomes the shortcomings caused by the absence of ultraviolet and visible light absorption characteristics of pentanediamine.C18 column separation,methanol-water for elution and evaporative light detector for detection,external standard quantification.By this method,the separation and content determination of pentanediamine and its raw material L-lysine can be realized within 10 minutes.The resolution of these two substances can reach more than 2,the linear range of pentanediamine is(94.5~1890)μg/mL,the linear range of L-lysine is(95~1890)μg/mL,the correlation coefficient(r)is 0.9992~0.9999,the linear relationship is good,and the spike experiments of three different concentration levels show that the recovery rate of pentanediamine is 97.05%~100.16%,and the RSD value is 0.42%~1.46%.This method is simple to operate,has high sensitivity,good accuracy and high test efficiency,and is suitable for analytical detection in the process of biosynthesis of pentanediamine and nylon 56.
