低共熔溶剂优化芹菜中芹菜素提取工艺研究

为优化芹菜素提取工艺条件,以芹菜素提取量为指标,采用单因素试验考察了 10种低共熔溶剂(Deep Eutectic Solvent,DES)及其氢键受体与氢键供体摩尔比、DES含水量、料液比、提取时间及提取温度等关键因素.在单因素试验基础上,通过Box-Behnken响应面法进一步优化芹菜素提取工艺.结果表明芹菜素最佳提取工艺为:氯化胆碱/乙酸摩尔比为1∶4,DES含水量为 30%,料液比为1∶38 g/mL,提取时间为2h,提取温度为50℃,此条件下芹菜素提取量为16.98 mg/g,较传统溶剂(乙醇)对芹菜素的提取量有明显提升,说明DES可作提取芹菜素的有效溶剂,可为芹菜素的提取及开发提供参考.
Optimization of Extraction Process of Apigenin in Celery by Deep Eutectic Solvent
In order to optimize the extraction of apigenin,10 types of deep eutectic solvents(DESs)and their molar ratio of hydrogen bond acceptor to hydrogen bond donor,water content of DES,solid-liquid ratio,extraction time and extraction temperature were investigated by single factor experiment with the extraction amount of apigenin as the index.Based on the single factor experiment,the extraction process of apigenin was further optimized by Box-Behnken response surface method.The results showed that the optimum extraction of apigenin was:the molar ratio of choline chloride and acetic acid,1∶4;water content of DES,30%;the ratio of material to liquid,1∶38(g/mL);extraction time,2 hours.extraction temperature,50℃.Under the conditions,the extraction amount of apigenin was 16.98 mg/g,which was significantly higher than that of traditional solvent(ethanol),indicating that DES could be used as an effective solvent for the extraction of apigenin,which could provide reference for the extraction and development of apigenin.
