目的:探究芍药苷-6'-O-苯磺酸酯在甲醇中的降解杂质,确证其结构,阐明降解机理。方法:将芍药苷-6'-O-苯磺酸酯溶于甲醇中,在加热的条件下生成降解杂质,通过硅胶柱层析和制备液相分离得到2个降解杂质,采用核磁共振、红外、质谱等技术确证杂质的结构,并对其降解机理进行推理。结果:鉴定了 2个降解杂质结构,推测出芍药苷-6'-O-苯磺酸酯在甲醇中的降解机理。结论:为芍药苷-6'-O-苯磺酸酯的杂质研究提供了物质基础,同时也为其工艺路线的选择、分析方法开发以及质量研究与控制提供较为科学的依据。
Structure Identification and Mechanism Analysis of Degradation Impurities from Paeoniflorin-6'-O-benzene Sulfonate in Methanol
Objective:To study the structure of degradation impurities and degradation mechanism of paeoniflorin-6'-O-benzene sulfonate in methanol.Methods:The paeoniflorin-6'-O-benzene sulfonate was dissolved in methanol to form degradation impurities under heating conditions,then the two impurities were isolated by column chromatography and preparative HPLC.Their structures were confirmed by NMR,IR and MS,and the degradation mechanism was deduced.Results:Structure of two impurities of paeoniflorin-6'-O-benzene sulfonate in methanol were identified and the degradation mechanism was deduced.Conclusion:This study provides the material basis for the impurity research of paeoniflorin-6'-O-benzene sulfonate,as well as the scientific basis for the selection of the technological process,the development of analytical methods and the quality control.
