Preparation of Lycium Barbarum Functional Peptides by Co Immobilization of Composite Proteases based on Cross-linked Enzyme Aggregates
In this study,co-immobilization composite proteases was prepared by the cross-linked enzyme aggregates.With activity recovery as the index,the maximal activity recovery of the cross-linked composite protease aggregates was 80.32%by single factor experiment under the optimal preparation conditions including:ratio of flavor protease to papain was 3∶1,precipitant((NH4)2SO4)concentration was 60%,glutaraldehyde concentration was 0.5%,crosslinking time was 50 min,and the crosslinking pH was 7.5.Compared with the single protease,the cross-linked composite protease aggregates showed better thermal and pH stability,and exhibited better performance of continuous hydrolysis of Lycium barbarum protein.Using proteolytic degree as index,the optimal hydrolysis conditions of the cross-linked composite protease aggregates were as follows:temperature 60 ℃,pH 6,enzyme concentration 10 mg/mL and hydrolysis time 100 min.Under these conditions,the maximal proteolytic degree reached 41.21%,which was 6.9%higher than that of free composite proteases.
