Study on the Standard Decoction of Polygoni Multiflori Caulis from Different Origins based on Fingerprint and Chemometrics
Objective:To explore the different components of Standard decoction of Shouwuteng(Polygoni Multiflori Caulis)from different producing areas by fingerprint combined with cluster analysis and other statistical methods,so as to provide a basis for the formulation of quality standards.Methods:The common peaks of 20 batches of Wuteng standard decoction were calibrated by the best fingerprint sample preparation method obtained by pre-treatment,and then the similarity of common peaks was evaluated combined with the"Traditional Chinese Medicine chromatographic Fingerprint Similarity Evaluation System"(2012 edition).Finally,the differential markers in different regions were screened by chemical pattern recognition method.Results:20 batches of Shouwu Teng standard decoction contained 8 common peaks,and their similarity evaluation values were all above 0.85.A total of 4 chromatographic peaks were identified.The 20 batches of samples were divided into four groups by Hierachical Cluster Analysis(HCA)and Principal Component Analysis(PCA),and the samples from the same place of origin were in the same group.According to partial least squares discriminant analysis(OPLS-DA)analysis and screening,the main differential markers in different regions were peak 1(2,3,5,4'-tetrahydroxystilbene-2-O-beta-D-glucoside),6,7(emodin)and 8(emodin methyl ether).Conclusion:The fingerprint method established in this study and the study of differential markers can provide reference for the improvement of the quality standard of Shenwu Decoction and its preparations.
