Preparation of Polysaccharides from Stachys Sieboldii and their Effect on High Glucose-induced Rat Glomerular Mesangial Cells
In order to study the effect of the polysaccharides from S.sieboldii on high glucose-induced glomerular mesangial cells(HBZY-1)in rats.The aqueous extract of S.sieboldii was deproteinised and separated by AB-8 macroporous resin column chromatography to obtain the crude polysaccharides of S.sieboldii,and then subjected to graded alcoholic precipitation to obtain the 30%alcoholic precipitation fraction(SSMP-30%),60%alcoholic precipitation fraction(SSMP-60%)and 90%alcoholic precipitation fraction(SSMP-90%),in which SSMP-90%had the highest content.The in vitro activity of the alcohol precipitated fractions was tested using high glucose-induced HBZY-1 as a cell model.The results showed that the different alcohol precipitation fractions were non-toxic to the model cells,and compared with the high glucose control group,the SSMP-30%and SSMP-90%fractions had a significant inhibitory effect on the proliferation of high glucose-induced HBZY-1(P<0.01)in a concentration-dependent manner,in which SSMP-90%had the strongest activity.It was suggested that polysaccharides from S.sieboldii had a protective effect on high glucose-induced glomerular mesangial cells in rats.
